The tissues I introduced in march are doing really good, now it is like the 4th medium change, but I do not get them to make callus, any idea which could be a good medium to induce a shoot to make a callus?

hi coline sorry for the late reply for callus i use 1/3 MS with full micronutrients,0.2mg/liter BAP and 0.1mg/liter of TDZ work well

bananaman i did the usual thing and searched the net if you look at a few of the mushroom growing web sites/forums they will help a lot once you get the basic principal and how it needs to work the rest is easy

hi coline sorry for the late reply for callus i use 1/3 MS with full micronutrients,0.2mg/liter BAP and 0.1mg/liter of TDZ work well I had a really bad callus using about 2mg BAP on N. x gentle, that formed at the base of the plant, it took all the nutrients from it and did not even make a single shoot from it, also on king tiger x mirabilis kuraburi happened that. So now I'm using only 0,5NAA and the plants seem to grow faster than ever, letting me make "in vitro nepenthes cuttings" and make many more. If you remember I started with nepenthes tissue on my very first attempt, and those were only 2 shoots, now I have more than 20 in a single year, and growing, just don't have as much time as I'd like to propagate all of them (including 400 of the other nepenthes, orchids, sarracenia, helis...)

Wow, the TDZ is incredible!, I got some and tried it on the plants, and my main 3 species in vitro in just 15 days have made a shoot growth from each leaf axil!
