Is your sterilization protocol the same for explant tissues? Do you find that 50% MS burns explants as well, this is what ive been using, with no positive results. I may be sterilizing too harshly, though. You mentioned Ampullaria seed as needing longer sterilization, any others? Thanks for all the info!

for some reason when i TC ampullaria seed it always has contamination so i give it a lot longer in the PPM about 24 hours even this does not always work. all other nep seed seems to respond to about four hour. i never have much luck with amp seed i have been told that it has a very short life only a few weeks. For ex-plants i use alcohol dip rinse with water and then a peroxide soak for two or three minutes then another rinse in water then a soak in PPM the same as for seeds. I don't have a great record with nepenthes ex-plants i have just about given up. I will be Trying a few more experiments with nepenthes meristems over the next few months hope i have better results this time

Robert, thanks for all the help. When you replate nepenthes that are established (ie small roots/shoots/pitchers develop) what media do you use to encourage root growth, and multiplication ? I have a few Dionaea on standard 50% MS, no hormones, they really need to move to new vessels what do you recommend for root growth and multiplication? Thanks in advance!

I have always used 0.5ml of NAA per liter of media but i have started to cut out the NAA and just use plain media as plants tend to form roots without the NAA but in saying this the roots that form when using NAA are much healthier than the ones in just plain media so i may go back to using NAA. plants grown with NAA tend to be more robust when hardening off As for shoot growth all i do is remove the callus that i have grow and place it into plain media and allow it to start growing shoots when i think that it has grown enough shoots i divide this clump up and place plants into media to grow shoots i also remove some of the left over callus and place it into media containing BAP to allow it to grow some more

Step by step guide i use a well grown plantlet place this plant in media containing 3-4ml BAP per liter of media. Once it has started to form some callus i cut the growing callus from the plant then place the callus into the same media This is the callus after i have removed it from the plantlet After it has grow to a large size i remove it from the media cut it into several sections removing the older and dead parts so i only place strong growing callus into the next stage callus has been placed into plain media (no BAP) allowing shoots to form I also place some callus back into media containing BAP to grow more callus callus has grown shoots i move this callus into bigger containers as it out grows the one it is in shoots cut from the callus and placed into media containing 0.5ml of NAA per liter of media if youi can leave a small amount of the callus on the shoots this seems to grow roots faster shoots have grow roots and are ready to be remove from TC and hardened off

I use a product called Growgel i buy it from a local suppler but it comes in a plain bag so i dont know if it has another product name.

Be carefull with moving nep's into callus. It can cause mutations easier than other propagation techniques.

Hi Robert i had successful germination in-vitro ampullaria seeds in 22 days. I would say the seeds you gotten were not fresh.

Hi Robert, yes ampullaria seed has only a few weeks of shelf life. I have managed to start several different forms of ampullaria seed lately. Here are a few pic. Amp tri colour form ampullaria red Ampullaria giant red Ampullaria harlequin ampullaria giant

Hi everyone here are a few photos of my latest batch of nepenthes in TC. this is a plant grown from seed and placed into media with 4ml of BAP. plant will start to form a callus then i move it into media with 2ml BAP and 0.2 ml of NAA, this will cause the callus to start growing shoots Once it has out grown the tube i move it to a larger container with plain media and let it grow plants will some times start to form roots. plant is still in same plain media Plant has filled the container so time to remove and seperate plants Plant has been divided up and placed into media containing 0.5ml of NAA I try to leave some callus on the plants this will help with root growth but roots will form without any callus Final result plant has been hardened off for two months and has formed its first descent pitcher
